Tobacco pesticide residue rapid detecting card

1. Using method

1.1 Sample pretreatment

For flesh tobacco:

Take 2g sample and cut them into small pieces (about 1cm²). Then put them in the beaker, and add 10ml pure water. Shake it evenly and stand it for 5mins.

Add 1ml pure water to chromatography column A, and use needle tubing to increase the pressure to drain the water. Suck 1ml above soak solution into chromatography column, and use needle tubing to increase the pressure slowly. Get rid of the first drop of solution and collect colorless clean filtered solution as testing solution. Drip 2 drops (namely 80ul) of testing solution on the white pill.

For the dehydrated and basked tobacco:

Take 2g sample and cut them into small pieces (about 1cm²). Then put them in the beaker, and add 40ml pure water. Shake it evenly and stand it for 5mins. Add 1ml pure water to chromatography column B, and use needle tubing to increase the pressure to drain the water. Suck 1ml above soak solution into chromatography column, and use needle tubing to increase thepressure slowly. Get rid of the first drop of solution and collect colorless clean filtered solution as testing solution. Drip 2 drops (namely 80ul) of testing solution on the white pill.

1.2  Reaction

Place the detecting card in the incubator at 40℃ for 10mins for pre-reaction. Keep temperature consistent for 3mins after pre-reaction to superimpose red pill on white pill for reaction. Judge the result according to the color variation of the white pill.

Do a blank control test with pure water or distilled water.

2. Result

Compared with blank control test, it is positive result that the white pill does not change the color or it just turns light blue. It indicates there is a high pesticide residue that the white pill does not change the color. And it indicates there is a relatively low pesticide residue that the white pill turns light blue. While it is negative result that white pill turns blue or has the same color with blank control test card. For those samples of which the results are positive, use other analysis methods to determine the types and concentration of the pesticide.

4. Matters need attention:

4.1 Shake the reaction tubes evenly and avoid shaking acutely.

4.2 Control the speed during using needle tubing to increase the pressure of chromatography column and don’t increase too hard in case collect deeper color filtering liquid which will disturb the result.

4.3 Tobacco pesticide detecting card is very sensitive to pesticide. The result will be disturbed if there are pesticides or safety insecticides around, or the operators or utensils are attached a small quantity of pesticide.

4.4 The time of white pill superimposing red pill is 3mins. Blue color will be deeper after 3mins, and the color will fade away after 24 hours.

5. Preservation condition:

This product should be stored in dry and dark condition. And it should be kept in the refrigerator at 4℃. And the expiration time is one year.
